A psychrotolerant Pseudomonas sp. AKM-L5 isolated from soil of Jammu city, India was investigated for the production of cold active lipase (CALip5). Further CALip5 was purified 120.14-fold by Sephadex G-100 ion-exchange chromatography and its molecular mass was estimated to be 57kDa by SDS-PAGE. The Km and Vmax values were found to be 0.108mg/mL and 542.59 U/mg, respectively. CALip5 was more active at 10oC and pH 7.0 whereas more stability was observed at 5oC and pH 8.0. CALip5 activity was significantly stimulated by 50mMCu2+ and various organic solvents whereas EDTA had very less effect. Cold temperatures washing performances were done and maximum percentage of oil was removed by the CALip5 than other detergents. High stability and activity at cold temperature in presence of organic solvents and metal ions and high oil removal capability make CALip5 a novel candidate for industrial application.
