<P><B>Abstract</B></P><P>The morphology, cultural characteristics and internal transcribed spacer’s (ITS)-5.8S rDNA analysis of TP-02 were applied to identify a strain of filamentous fungus <I>Rhizopus</I> TP-02 with high activity of cellulase. The result of that showed that the novel strain could be identified as <I>Zygomycotina</I>, <I>Phycomycetes</I>, <I>Mucorales</I>, <I>Mucoraceae</I>, <I>Rhizopus stolonifer</I> var. <I>reflexus</I>. The filter paper activity and CMC activity of the strain TP-02 could achieve 7.56 IU/mL and 28.5 IU/mL respectively after 96 h fermentation. During the tests of purification procedure, three types of Endo-β-1, 4-glucanases and two different molecular weight of β-1, 4-glucosidases were detected from the crud enzyme produced by the strain.</P> <P><B>Highlights</B></P><P>► A novel strain <I>Rhizopus stolonifer</I> var. <I>reflexus</I> was well identified and characterized. ► The <I>stain</I> is very active to produce the cellulase during fermentation characteristics. ► The enzyme characteristics of the cellulose were also carried out.</P>
